Er stammte aus dem <a href="Assen_(Adelsgeschlecht)" title="Assen (Adelsgeschlecht)">Hause Assen</a>. Mizo Assen war mit <a href="Maria_Assenina" title="Maria Assenina">Maria Assenina</a>, eine Tochter des Zaren <a href="Iwan_Assen_II." title="Iwan Assen II.">Iwan Assen II.</a> verheiratet, mit der er einen Sohn, <a href="Iwan_Assen_III." title="Iwan Assen III.">Iwan Assen III.</a> und eine Tochter Kira-Maria Assenina hatte.
